Treatment protocol |
We use shRNA to knock down(KD) the expression of LKB1 gene at the human acute myeloid leukemia KG1a cells ,meanwhile build the negative control with normal LKB1 expression.

Growth protocol |
KG1a cells were maintained in Iscove’s modified Dulbecco’s media supplemented with 10% fetal bovine serum. All of the cells were incubated at 37°C in a humidified atmosphere with 5% CO2.

Trizol extraction of total RNA was performed according to the manufacturer's instructions.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45°C on GeneChip® PrimeView™ Human Gene Expression Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the GeneChip Scanner 3000.

Data processing |
The data were analyzed with rma using Affymetrix default analysis settings and global scaling as normalization method.
