Extraction protocol Total RNA was prepared from individual tissue samples using Tri-Reagent isolation protocol. Three pools (6 animals) were prepared for each experimental group.

Hybridization protocol The membranes were prehybridized for 1 h at 68oC in a hybridization solution (Clontech) containing 0.1 mg/ml sheared salmon testes DNA (Sigma) to block nonspecific binding. Synthesized radio labeled cDNA probe was applied to each membrane and hybridized overnight at 68oC. Cot-1 DNA (Clontech) (1 _g/ml) was added to block nonspecific binding.
Scan protocol For analysis, the membranes were exposed for 15 and 24 h to a phosphor screen and detected using a Bio-imaging Analyzer BAS2000 (Fuji Photo Film Co. Ltd Japan.
Description left ventricle of heart excised from rats subjected to heat stress at 41oC (2h followed by 1h recovery at 24oC, following heat acclimation (34oC, 1mo).
Data processing We used Atlas Image 2.01 software (Clontech) to grid the phosphor image, to record the pixel density of each spot, and to perform background subtraction. The background-subtracted data were then further analyzed.
