Summary |
To dissect the role of 17a-estradiol in lifespan extending and its potential side effects for long-term administration, the pooled hypothalami from aging male Norway brown rats treated with 17a-estradiol for 6 months (O.T), aged male control (O), and young male control (Y) were subjected to single-nucleus transcriptomic sequencing (snRNA-seq).

Overall design |
24-month-old-male Norway brown rats were fed freely with diet mixed with 17a-estradiol at a dose of 14.4 milligrams per kilogram diet (14.4 ppm) starting at 24 months of age for six months (O.T). 30-month-old-male rats and 4-month-old-male rats were used as aging control (O) and young sample control (Y) respectively. All rats had ad libitum access to food and water. The intact hypothalami in each group were pooled for nucleus isolation and snRNA-seq.
