Title Gain-of-function genetic perturbationssimultaneouslyacross500 barcoded cancer cell lines
Organism Homo sapiens
Experiment type Other
Summary Using thisapproach, wequeriedthe pan-cancer vulnerability landscape upon activating 10key pathway nodes, revealing selectiveactivation dependenciesof MAPK and PI3K pathwaysassociatedwith specific biomarkers

Overall design To profile activation of the curated gene sets across a broad panel of cell lines, we developed a high-throughput method for systematically examining the growth-suppressive effect of ORF expression in ~500 cancer cell lines. We next tested the perturbation effects of 11 ORFs (including GFP as a negative control) by lentiviral infection of each ORF encoding virus into separate pools of 488 cell lines.
