Somatic cells in testes

(Submitter supplied) Gene expression profiles of somatic cells of fetal and adult testis. To identify genes developmentally regulated in the somatic cells of the testis, serial analysis of gene expression (SAGE) has been used to generate gene expression profiles from these cells in the fetal and adult mouse. To avoid germ cell transcripts, a fetal SAGE library was generated from germ cell-free fetal Wv/Wv mice, and an adult SAGE library was generated from adult testes depleted of germ cells with busulfan. more...

CEA_SGF:E00004#TetOFF_Stat3F

(Submitter supplied) The goal of this study was to identify the target genes of Stat3 involved in murine ES cell self-renewal. To achieve this goal, we used the Gs2 ES cell line expressing an inducible dominant negative mutant of Stat3 (Stat3F) in wich Y705 is mutated to phenilalanine. The Gs2 ES cells are routinely maintained in the presence of LIF 1000 U/ml and tetracycline 1µg/ml (LIF/ON). In this condition, Stat3F is not expressed and the cells are maintained in an undifferentiated phenotype. more...

CEA_SGF:E00005#LIF

(Submitter supplied) In order to identify the genes involved in LIF/gp130 self-renewal response, we analyzed the transcriptome of Gs2 ES cells induced to differentiate upon LIF withdrawal for 16h, 24h and 48h (LIF16, LIF24 and LIF48 respectively)and compared them to that of undifferentiated ES Gs2 cells continuously maintained in the presence of LIF. Since this cell line also carry a tetracycline regulatable dominant negative form of Stat3 (Stat3F, all the experiment was done in the presence of Tetracycline (tet ON) to avoid Stat3F expression. more...

Global transcription analysis of immature avian erythrocytic progenitors

(Submitter supplied) The molecular mechanisms regulating the cell fate decision between self-renewal and differentiation/apoptosis in stem and progenitor cells are poorly understood. Here, we report the first comprehensive identification of genes potentially involved in the switch from self-renewal toward differentiation of primary, non-immortalized erythroid avian progenitor cells (T2EC cells). We used the Serial Analysis of Gene Expression (SAGE) technique in order to identify and quantify the genome fraction functionally active in a self-renewing versus a differentiating cell population. more...

Apoptosis and differentiation commitment:novel insights revealed by gene profiling studies in mouse Embryonic Stem cells

(Submitter supplied) Mouse embryonic stem (ES) cells remain pluripotent in vitro when grown in presence of Leukaemia Inhibitory Factor (LIF). LIF starvation leads to apoptosis of some of the ES-derived differentiated cells, together with p38a MAP kinase activation. Apoptosis, but not morphological cell differentiation, is blocked by a p38 inhibitor, PD 169316. To further understand the mechanism of action of this compound, we have identified its specific targets by microarray studies. more...

Embryonic fibroblast-independent embryonic stem cell differentiation

Analysis of CGR8 embryoid bodies differentiating for 3 or 10 days. Culturing of most embryonic stem (ES) cells require embryonic fibroblast (MEF) feeder cells which may affect their gene expression profiles. CGR8 cells are MEF-independent and represent a powerful model of ES cell differentiation.

Juvenile idiopathic arthritis: peripheral blood cells

(Submitter supplied) Systemic-onset juvenile idiopathic arthritis (soJIA) is a rheumatic disease in childhood characterized by systemic symptoms and a relatively poor prognosis. The peripheral leukocytes are thought to play the pathological role of soJIA although the exact cause is still obscure. In this study, we aimed to clarify the cellular functional abnormality in those cells. Here, we analyzed the gene expression profile in peripheral leukocytes from 51 patients with soJIA, 6 patients with poly-articular type JIA (polyJIA) and 8 healthy children utilizing DNA microarrays. more...
