Developmentally regulated alternate 3’ end cleavage of nascent transcripts controls dynamic changes in protein expression in an adult stem cell lineage

(Submitter supplied) To determine the extent of alternative 3' end cleavage during Drosophila spermatogenesis, we performed 3' end sequencing of Drosophila testes from flies at different time points post heat shock (pHS) that were mutant for bam and also had a heat shock inducible transgene expressing Bam. We then performed Polysome profiling of testes at 24, 48 and 72 hours PHS and created 3' end sequencing libraries from the free, 40S, 60s, 80S, 2-3 ribosomes, and 4+ ribosomes fractions for each dataset

Identifying the transcriptomic effects of tRNA-induced proline-to-serine mistranslation in Drosophila melanogaster

Mistranslation has diverse effects on multicellular eukaryotes and is implicated in several human diseases. Tudor domain containing protein 5-like(Tdrd5l) identifies a novel germline body and regulates maternal RNAs during oogenesis

(Submitter supplied) In this study we show that Drosophila Tudor-domain containing protein 5-like (Tdrd5l) is important for post-transcriptional regulation of maternal mRNAs. Tdrd5l localizes to a novel of body in the germline and interacts with post-transcriptional regulation pathways. In mutant females we observe accumulation of Gurken and its activator Orb in nurse cells during oogenesis. Decreased orb function suppresses the Tdrd5l-mutant phenotype, indicating that this is a primary defect in these mutants.

Linker histone H1 regulates homeostasis of heterochromatin associated cRNAs [ChIP-Seq 3]

(Submitter supplied) Chromosomal RNAs (cRNAs) are a poorly understood fraction of cellular RNAs that co-purify with chromatin. Here we show that, in Drosophila, cRNAs constitute a heterogeneous group of RNA species that cover ~28% of the genome. Intriguingly, we found that cRNAs are highly enriched in heterochromatic transcripts.
