Major bacterial pathogens in the United States and around the world, including Salmonella enterica, E. coli, Shigella, Listeria monocytogenes, Campylobacter Jejuni, Vibrio parahaemolyticus. This project also contains WGS from many food-associated organisms that have unique traits.

Construction protocol: SMRTbell sequencing libraries were generated using standard procedures (version C2; http://www.pacb.com/samplenet/PC_10kb_Template_Preparation_and_Sequencing.pdf). gDNA was sheared to an average size of approximately 10 kb using g-TUBEs (Covaris). Fragmented DNA underwent DNA Damage Repair, End Repair, and blunt hairpin adapters were ligated onto each end. Incompletely formed SMRTbell templates were degraded with a combination of Exo III and Exo VII. SMRTbell templates were magbead loaded and sequenced with C2 chemistry.
