Nonfucosylation of an anti-TIGIT antibody enhances FcyR engagement, driving innate immune activation and antitumor activity

(Submitter supplied) TIGIT is an immune checkpoint receptor expressed on activated and memory T cells, immunosuppressive T regulatory cells, and natural killer (NK) cells. TIGIT has emerged as an attractive target for antitumor therapies, due to its proposed immunosuppressive effects on lymphocyte function and T cell activation. We generated an anti-TIGIT monoclonal antibody (mAb) that binds with high affinity to human, non-human primate, and murine TIGIT and through multiple experimental methodologies demonstrated that checkpoint blockade alone is insufficient for antitumor activity. more...

Single Cell RNA-seq on Human PBMC

(Submitter supplied) To assess the impact of IgG1 and IgG4 variants of a TIGIT-Fc-LIGHT fusion protein on human PBMC expression, each fusion protein or an untreated control were incubated with human PBMC for 2 days in AIMV lymphoproliferative media. Both IgG1 and IgG4 variants of TIGIT-Fc-LIGHT induced broad immune cell activation on T, NK, and myeloid cells.

Anti-TIGIT antibody improves PD-L1 blockade through myeloid and Treg cells

(Submitter supplied) Tiragolumab, an anti-TIGIT antibody with an active IgG1κ Fc, demonstrated improved outcomes in the phase 2 CITYSCAPE trial (ClinicalTrials.gov: NCT03563716) when combined with atezolizumab (anti-PD-L1) versus atezolizumab alone. However, there remains little consensus on the mechanism(s) of response with this combination. Here we find that a high baseline of intratumoural macrophages and regulatory T cells is associated with better outcomes in patients treated with atezolizumab plus tiragolumab but not with atezolizumab alone. more...

Genome-wide chromatin accessibility changes in cultured primary human monocytes cultured at low density and high density.

(Submitter supplied) ATAC sequencing of primary human monocytes cultured at low and high density. Monocytes isolated from PBMCs of 3 healthy donors were cultured at low density (1 x 10^6 cells/mL) or at high density (1 x10^7 cells/mL) for 24hrs and harvested for ATAC sequencing. Protein expression of FcgR2b is higher on monocytes in high density conditions compared to low density conditions, where expression is negligble. more...

Transcriptome-wide expression changes over time in cultured primary human monocytes due to DMOG treatment

(Submitter supplied) RNA sequencing of primary human monocytes cultured with and without hypoxia-mimetic agent DMOG (Dimethyloxalylglycine). Monocytes isolated from PBMCs of 7 healthy donors were cultured with and without DMOG for 24hrs and harvested at 0, 2, 10 or 24hrs for RNA sequencing. DMOG suppresses HIF Prolyl hydroxylase (HIF-PH) activity by acting as a small molecule competitive inhibitor. HIF-PH inhibition leads to increase in endogenous HIF protein levels and mimics aspects of hypoxia. more...

Efficacious anti-CTLA-4 antibodies drive myeloid activation and TME reprograming through FcγR engagement and Type-I interferon signaling

(Submitter supplied) Despite the clinical success of checkpoints inhibitors, a significant gap still exists in our understanding of their in vivo mechanism of action, thus limiting the rational development of next generation agents. Amongst these, anti-CTLA-4 antibodies were originally developed to block inhibitory signals into activated effector T cells. However, several recent studies have demonstrated that depletion of regulatory T cells expressing high levels of CTLA-4 is critical to anti-CTLA-4 anti-tumor activity. more...
