Hepatitis C virus RNA is 5’ flavin adenine dinucleotide (FAD) capped

(Submitter supplied) RNA viruses have developed elaborate strategies for 5’ capping and protection of their genomes. However, so far no 5’ RNA cap has been identified for Hepatitis C virus (HCV), which cause chronic infection, liver cirrhosis and cancer in humans4. Here, we demonstrate that the cellular metabolite flavin adenine dinucleotide (FAD) is used as noncanonical initiating nucleotide by the viral RNA-dependent RNA polymerase resulting in a 5’ FAD cap on the HCV RNA. more...

High-throughput single-cell RNA-Seq profiling of DMSO-differentiated Huh7.5.1 undergoing or not long-term HCV infection

(Submitter supplied) Hepatocellular carcinoma (HCC) is the second leading cause of cancer death worldwide, indicating urgent need for novel preventive strategies. Cancer chemoprevention discovery has been challenging due to the absence of tractable and clinically relevant model systems. Here, we developed a simple and robust human liver cell-based system, in which persistent hepatitis C virus (HCV) infection induces a HCC risk signature robustly predicting long-term HCC risk in cirrhotic patients. more...

The Microbial Detection Array for Detection of Emerging Viruses in Clinical Samples - A Useful Panmicrobial Diagnostic Tool--Array Version 2f

(Submitter supplied) Emerging viruses are usually endemic to tropical and sub-tropical regions of the world, but increased global travelling, climate changes and changes in lifestyle are believed to contribute to the spread of these viruses into new regions. For many of them, the disease symptoms are similar to each other, as well as to other more common diseases, making them difficult to diagnose. A rapid identification will help to decide about specific treatment and appropriate case management. more...

Therapeutic silencing of microRNA-122 in primates with chronic hepatitis C virus infection

(Submitter supplied) The aim of this deep sequencing project is to determine whether treatment of chronically Hepatitis C infected chimpanzees with an antiviral compound leads to accumulation of viral mutants. The liver-expressed microRNA-122 (miR-122) is essential for hepatitis C virus (HCV) RNA accumulation in cultured liver cells, but its potential as a target for antiviral intervention has not been assessed. Here, we show that treatment of chronically infected chimpanzees with a locked nucleic acid (LNA)-modified oligonucleotide (SPC3649) complementary to miR-122 leads to long-lasting suppression of HCV viremia with no evidence for viral resistance or side effects in the treated animals. more...

PEPperCHIP Autoimmunity Microarray 2.0

(Submitter supplied) Laser printer spotted peptides Protocol: Amino acid microparticles are laser printed onto a glass slide with a custom 24-color laser printer. One layer of an amino acid particle pattern is printed on a coated glass slide. A heating step melts the particles. Embedded amino acids can couple to the chip surface. In accordance with solid phase Fmoc chemistry, a cycle of synthesis is completed after removal of excessive material by washing and cleavage of the N-terminal Fmoc protection group. more...
